站房主体为两层，两侧三层，总建筑面积14984平方米。建筑主体高度16.2~28.2米，总长164米，总宽44米。

徐州东站位于徐州火车站以东10公里，京福高速公路以西。

徐州东站为徐州地铁1号线，徐州地铁6号线和徐州地铁7号线换乘为徐州地铁一号线一期工程的东终端站。1号线车站位于徐州东站楼南侧，6号线车站位于东站楼地下一层。该站紧邻徐州东站和车站广场，南有绿地窗口，徐州东长途汽车站和正在建设的徐州东站东站场。
